چکیده مقاله:

Cultivar of roses (Rosa spp.) has been grown in greenhouses in Qazvin region of Iran for local markets. Agrobacterium tumefaciens strains were isolated and identified from six different samples of roses plants imported from the Netherland to Iran. During August and September of 2012, nearly 2-5% of rose plants in two different greenhouses in the province of Qazvin were observed with crown gall symptoms on various parts, particularly in the stem. A non-fluorescent gram-negative bacterium was consistently isolated from diseased tissues on to King's B medium. The isolated strains were confirmed asAgrobacterium using different biochemical and pathogenicity (tumor forming ability) tests. Four representative strains isolated were aerobic, non-sporing, non-pigmented, motile, rod-shaped, oxidase negative and catalase positive. All of four isolates were positive for tumor forming ability in pathogenicity tests on carrot disc and tomato stem.

Plant responses to Agrobacterium tumefaciens and crown gall development

Agrobacterium tumefaciens causes crown gall disease on various plant species by introducing its T-DNA into the genome. Therefore, Agrobacterium has been extensively studied both as a pathogen and an important biotechnological tool. The infection process involves the transfer of T-DNA and virulence proteins into the plant cell. At that time the gene expression patterns of host plants differ depe...

Historical account on gaining insights on the mechanism of crown gall tumorigenesis induced by Agrobacterium tumefaciens

The plant tumor disease known as crown gall was not called by that name until more recent times. Galls on plants were described by Malpighi (1679) who believed that these extraordinary growth are spontaneously produced. Agrobacterium was first isolated from tumors in 1897 by Fridiano Cavara in Napoli, Italy. After this bacterium was recognized to be the cause of crown gall disease, questions we...

Agrobacterium tumefaciens Site Attachment as a Necessary Prerequisite for Crown Gall Tumor Formation on Potato Discs.

The infectivity of Agrobacterium tumefaciens strain B6 was inhibited about 50% when these bacteria were inoculated on potato discs with equal viable cell counts of a weakly virulent strain of A. tumefaciens (B-48) or autoclaved strains of B6 or B-48. Inhibition by B-48 or autoclaved B6 could still be obtained when these cells were added up to a maximum of 10 minutes after the addition of viable...

Mechanisms of tumor induction in crown gall. I. Production and pathogenicity of spheroplasts of Agrobacterium tumefaciens.

The hypothesis that crown gall tumorigenesis requires the formation of spheroplasts was tested by comparing the tumorinducing ability of populations of both permanent and revertable spheroplasts of Agrobacterium tumefaciens with that of popula tions of the typical rod form of the bacterium. Permanent spheroplasts produced by serial transfers in medium containing 0.005 MD-methionine did not prod...

In Vivo Synthesis of Crown Gall-specific Agrobacterium tumefaciens-directed Derivatives of Basic Amino Acids.

Several kinds of primary sunflower (Helianthus annuus) crown gall tissues were established in tissue culture and then labeled in vivo with either [(14)C]arginine, [(14)C]histidine, [(3)H]lysine, or [(3)H]ornithine. Crown gall tissues incited by Agrobacterium tumefaciens strains that utilize octopine as a sole source of carbon or nitrogen for growth synthesized the four members of the N(2)-(1-ca...
